Margarito grew up in Tijuana, Baja California, where he began to train at boxing at a local gym in his early teens. During his relatively brief amateur career, he had a record of 18-3. At the age of 15, he turned pro, defeating Jose Trujillo, and shortly afterwards he won the fight against Victor Angulo by knockout, the first in his career, after which he was defeated by Victor Lozoya. He went on to score some more notable wins, including over Alfred Ankamah, Juan Soberanes Ramos, Buck Smith, Sergio Martínez and David Kamau. His net worth started to rise.

In 2001 Margarito fought against Daniel Santos for his WBO Welterweight title. However, the match had to be stopped as both fighters suffered severe injuries due to a clash of heads and had to be sent to hospital. After Santos vacated the title the following year, Margarito fought against Antonio Díaz, and.in the tenth round he defeated Diaz by knockout to become the WBA Welterweight champion. His popularity and net worth were boosted.

He later successfully defended the title against Daniel Ramirez and Andrew Lewis, before switching to the light middleweight division, beating Maurice Brantley in 2003. The following year he defended his welterweight title, defeating Hercules Kyvelos, but shortly after he suffered a defeat in a rematch against Santos for his WBO Light Middleweight title.

The year 2005 saw one of the best victories in Margarito’s career, beating the undefeated boxer Kermit Cintron and retaining his WBO welterweight title. A year later, he again retained the title in a match against Jaime Manuel Gómez, and several months later defeated Joshua Clottey, setting a Compubox all-time record of 1675 total punches thrown in a twelve-round bout. His consequent wins significantly contributed to his popularity and to his wealth as well.

Margarito lost his WBA title in 2007, being defeated by Paul Williams. The following year, he faced Cintron again, who had meanwhile won the IBF Welterweight title; he defeated Cintron by knockout and took both titles.

Refusing to enter a rematch against Clottey, Margarito vacated his IBF title, choosing to fight against the undefeated WBA Welterweight Champion Miguel Cotto, whom he defeated by technical knockout, retaining his WBA title. However, the same year he suffered a defeat against Shane Mosley by technical knockout in the ninth round, losing his WBA title. Prior to the bout, Margarito’s corner was caught tampering with his hand wraps, which eventually proved to contain substances similar to plaster of Paris. Both Margarito and his trainer Capetillo were suspended for a year, and as a result, his 2008 win over Miguel Cotto, as well as his other victories, were brought into question.

Returning to the ring in 2010, Margarito won against light middleweight contender Roberto García, taking the vacant WBC International Light Middleweight title. Later that year, he fought Manny Pacquiao for the WBC Light Middleweight title, suffering a loss and a fractured eye bone that required a surgery. The following year he was defeated by Cotto, suffering injury in the same eye again, which brought his career into doubt, and Margarito retired from professional boxing. However, in 2016 he returned to the ring, winning two fights, the second for the WBO–NABO light middleweight title against Ramon Alvarez.

Speaking about his personal life, the boxer married Michele Margarito in the late ’90s, but they divorced and in 2011 he married Lorena Margarito, with whom he has two children, and a step-child. The family resides in Tijuana, Mexico.
