Concerning his professional career, Rolle was selected 8th in the first round of the NFL draft in 2005 by the Arizona Cardinals, where he played his first four years. In his first season with the new franchise, he played all 16 games and made 87 tackles and an interception. In the 2011 regular season, Rolle recorded 96 tackles, and the Giants ended the season with a 9-7 record, qualifying for the playoffs thanks to a decisive victory over the Cowboys. At the beginning of 2012, Antrel played in Super Bowl XLVI, won against the New England Patriots 21-17, becoming for the first time NFL champion. The 2013 season, Rolle ended in 2nd place in the league with 6 interceptions, 98 tackles, 2 sacks and 12 deflected passes, all new personal records, being selected in the Second All-Pro team and called for his third Pro Bowl. He was also listed 72nd in the NFL Top 100 by his colleagues.
In 2015, Rolle signed a three-year contract with the Chicago Bears, but was injured and started only seven games, then being released. He subsequently announced his retirement in late 2016.
